ACQUIT Y UPLC 顆粒技術(shù)
制造 UPLC 顆粒比合成小顆粒需要更多的技術(shù),絕大多數(shù) HPLC 顆粒都不能保持在超高效液相色譜操作壓力(例如:15000 psi/1000 bar)條件下的機(jī)械穩(wěn)定性和結(jié)構(gòu)完整性。為什么顆粒的耐壓性質(zhì)如此重要呢?為了*實(shí)現(xiàn)亞 2 微米顆粒的高柱效,需要在更高的線速度下(亦即更高體積流速)下進(jìn)行日常分離,而亞 2 微米小顆粒在更高的線速度條件下會(huì)產(chǎn)生更高的操作壓力。沃特世創(chuàng)造了兩種高效耐壓的 UPLC顆粒:1.7 μm 亞乙基橋雜化顆粒(BEH)和 1.8 μm 高強(qiáng)度硅膠顆粒(HSS)。
*個(gè) ACQUITY UPLC 顆粒是 1.7 μm 亞乙基橋雜化顆粒(BEH),這是沃特世第二代雜化顆粒,它是 UPLC 技術(shù)的一個(gè)關(guān)鍵推動(dòng)力,共有 5 種不同的鍵合相:C18, C8, Shield RP18,Phenyl 和 HILIC。由于它是雜化顆粒,因此具有更寬的 pH 使用范圍(pH 1-12),能夠幫助分離科學(xué)家快速有效地開發(fā)方法。同時(shí),BEH 顆粒技術(shù)也用于 XBridge 系列的 HPLC色譜柱,有不同粒徑(2.5、3.5、5 和 10 μm)供選,因此分離方法能夠在 HPLC 和 UPLC 技術(shù)平臺(tái)實(shí)現(xiàn)無(wú)縫轉(zhuǎn)換。
